    Recombinant HHV-1 TK (Thymidine kinase) expression in E.coli requires the insertion of the target DNA fragment into an E.coli expression vector, routinely a plasmid vector, and the transferral of this vector into E.coli cells. The cells are then cultured and induced to express this TK (Thymidine kinase) protein. The cells are harvested by centrifugation, samples prepared and proteins detected by polyacrylamide gel electrophoresis and subsequent staining of the gel with Coomassie Brilliant Blue or silver stain or by immunoblotting. The TK (Thymidine kinase) protein expression in E.coli is highly scalable and can be adjusted from the bacterial colony to conical flasks for liquid cultures, to fermentation reaction chambers.

    TK is a protein coding gene that encodes thymidine kinase. According to some studies, TK may have the following features.
    The efficiency of translation of thymidine kinase mRNA increases when cells begin DNA replication. This shift may facilitate fine-tuning of cell cycle regulation of thymidine kinase activity at the post-translational level. Thymidine kinase (TK) has a key function in the synthesis of DNA. Thymidine kinase is involved in DNA synthesis. Serial thymidine kinase levels in patients with systemic breast cancer reflect response to systemic therapy.
