CRTAM

CRTAM(CD355)是免疫球蛋白超家族(Ig-SF)的新成员,同时也是nectin样蛋白超家族的成员。它作为细胞表面跨膜蛋白,在免疫系统的形成、应答和细胞交互中扮演重要角色。CRTAM在活化的人NK细胞、CD8+ T细胞和CD4+ T细胞子集上表达,参与细胞粘附、极性和增殖,并调节淋巴细胞功能,如促进TCD8+细胞在淋巴结内的粘附和保留。 CRTAM与多种疾病关联,包括炎症、哮喘、恶性间皮瘤和某些肿瘤疾病。CRTAM能够影响CD4+ T细胞的极化,促进CD4+ T细胞的进一步分化,并增强细胞因子的分泌能力。此外,CRTAM在免疫过程中对NK细胞杀伤肿瘤细胞起到关键作用,呈现出潜在的肿瘤靶向策略。CRTAM还可以与Necl-2相互作用,在体外促进NK细胞的细胞毒性和CD8+ T细胞的IFN-γ分泌,以及在体内NK细胞介导的对表达Necl-2的肿瘤的排斥反应。 CRTAM的表达受到严格的调控,主要表达于激活的免疫细胞上。在健康人的外周血单个核细胞(PBMC)中,CRTAM的表达量很低,但在哮喘病人的PBMC中高表达。CRTAM的启动子与IL-2一样,受到AP-1转录因子的调控。CRTAM激活后会刺激CD8+ T细胞分泌IFN-γ,并增强CD8+ T细胞和NK细胞的杀伤能力。
