SIRPA

信号调节蛋白α(SIRPα,也称为CD172a)是一类广泛表达的糖蛋白分子,属于免疫球蛋白超家族结构域。SIRPα在髓系造血细胞(如巨噬细胞、树突状细胞)和神经细胞表面均有表达;而其配体CD47,也称整合素相关蛋白(integrin-associated protein,IAP),广泛表达于几乎所有正常细胞的表面。SIRPα与其他细胞上的CD47结合后,可向巨噬细胞传递抑制性信号,抑制巨噬细胞对靶细胞的吞噬作用。因此,这条通路可用来识别己细胞(self)与非己细胞(non-self)。一些肿瘤细胞利用这个机制,高表达CD47来向巨噬细胞发出“别吃我(Don’t eat me)”信号,来进行免疫逃逸。作为一个热门的免疫抑制性信号通路,目前有多种靶向Sirpα和/或CD47免疫检查点的抗体设计策略, 包括全长抗体、F(ab)、与Fc融合的F(ab)等。
