疱疹病毒进入介体(Herpes virus entry mediator A,HVEM,又称TNFRSF14)是TNF受体超家族成员,广泛表达于APCs、内皮细胞和淋巴细胞,尤其是幼稚T细胞。HVEM与多种蛋白结合,包括LIGHT、TNF-β、BTLA和CD160。当HVEM与LIGHT或LTα结合时,传递共刺激信号,而HVEM与BTLA或CD160结合则触发共抑制信号。因此,HVEM通常被描述为取决于所接合配体的分子开关。研究发现,HVEM在一些肿瘤中过表达,并且显示肿瘤浸润T细胞呈BTLA阳性。此外,HVEM的高表达与显著较差的预后相关。肿瘤中HVEM上调代表了一种免疫逃逸机制,类似于肿瘤细胞过度表达PD-L1或PD-L2。根据转录组学分析,HVEM表达与免疫细胞浸润和微环境中的基质细胞有关。HVEM的表达还与PD-1、PD-L1、CTLA-4、LAG-3和VISTA的表达相关。因此,HVEM在免疫和炎症反应的调节中起着至关重要的作用,尤其是T细胞的激活。
