    • IHC image of CSB-PA850415LA01HU diluted at 1:400 and staining in paraffin-embedded human endometrial cancer performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a biotinylated secondary antibody and visualized using an HRP conjugated SP system.
    • Immunofluorescence staining of Hela cells with CSB-PA850415LA01HU at 1:133, counter-stained with DAPI. The cells were fixed in 4% formaldehyde, permeabilized using 0.2% Triton X-100 and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4°C. The secondary antibody was Alexa Fluor 488-congugated AffiniPure Goat Anti-Rabbit IgG(H+L).

    The product CSB-PA850415LA01HU is an IgG rabbit polyclonal GSDMA antibody suitable for the recognition of human GSDMA by ELISA, WB, IHC, and IF. The recombinant human GSDMA protein (64-172aa) is used to immunize the rabbit to generate the GSDMA polyclonal antibody. GSDMA antibody is available in the non-conjugated IgG antibody form. Its purity is over 95% using protein G purification.

    The GSDMA protein act as a regulator of cell death and inflammation. GSDMA is primarily expressed in epithelial tissues, where it has been shown to play a critical role in regulating immune responses and tissue homeostasis. It also participates in various physiological processes, including skin development and maintenance of the skin barrier function. Dysregulation of GSDMA causes skin disorders, inflammatory bowel disease, cancer, and other diseases.

    This form constitutes the precursor of the pore-forming protein: upon cleavage, the released N-terminal moiety (Gasdermin-A, N-terminal) binds to membranes and forms pores, triggering cell death.; Pore-forming protein that causes membrane permeabilization and pyroptosis. Released upon cleavage in vitro of genetically engineered GSDMA, and binds to membrane inner leaflet lipids. Homooligomerizes within the membrane and forms pores of 10-15 nanometers (nm) of inner diameter, triggering pyroptosis. Binds to membrane inner leaflet lipids, such as phosphatidylinositol (4,5)-bisphosphate. The functional mechanisms and physiological proteases that cleave and activate this pore-forming protein are unknown.
