The synthesis of this Recombinant Human TRIM72 protein depends on the utilization of recombinant DNA technology to a large degree. DNA sequences that encoded the TRIM72 protein could be inserted into a vector and introduced into an expression host, E.coli, where it could be easily expressed in and purified from. The expression of this TRIM72 protein was at 2-477aa. N-terminal GST tag was fused with this protein. The purity is 85%+ determined by SDS-PAGE.
TRIM72 (also known as MG53) is gene encoding a protein named tripartite motif-containing protein 72 (TRIM72) or mitsugumin-53 (Mg53) in human. The protein encoded by this gene is a muscle-specific TRIM family protein. This protein is initially identified as a key regulator of cell membrane repair machinery. It has been suggested to be a critical regulator of muscle insulin signaling pathway by acting as ubiquitin E3 ligase targeting both the insulin receptor and insulin receptor substrate 1 (IRS1).

功能:Muscle-specific protein that plays a central role in cell membrane repair by nucleating the assembly of the repair machinery at injury sites. Specifically binds phosphatidylserine. Acts as a sensor of oxidation: upon membrane damage, entry of extracellular oxidative environment results in disulfide bond formation and homooligomerization at the injury site. This oligomerization acts as a nucleation site for recruitment of TRIM72-containing vesicles to the injury site, leading to membrane patch formation. Probably acts upstream of the Ca(2+)-dependent membrane resealing process. Required for transport of DYSF to sites of cell injury during repair patch formation. Regulates membrane budding and exocytosis. May be involved in the regulation of the mobility of KCNB1-containing endocytic vesicles.
