The production of this recombinant S.viridochromogenes pat protein is just like all recombinant proteins. The process involved transfecting E.coli cells with DNA vector containing the template of recombinant DNA. The E.coli cells containing the template were then cultured so that they could transcribe and translate the pat protein. N-terminal 6xHis-SUMO tag was used in the process. The purity is 90% determined by SDS-PAGE.
PAT is a gene providing instructions for making a protein called phosphinothricin N-acetyltransferase (also known as PPT N-acetyltransferase or phosphinothricin-resistance protein). This protein, an effector of phosphinothricin tripeptide (PTT or bialaphos) resistance, has N-acetyltransferase and phosphinothricin N-acetyltransferase activity involved in response to antibiotic and herbicide. This enzyme inactivates phosphinothricin (PPT) through a transfer of an acetyl group from acetyl CoA.
