PDE4A
PDE4，即磷酸二酯酶4（phosphodiesterase 4），是磷酸二酯酶家族中的一个重要成员，由4个同源基因编码，产生4种亚型：PDE4A、PDE4B、PDE4C和PDE4D。PDE4主要在免疫细胞和多种炎症细胞中表达，特异性水解细胞内的第二信使环磷酸腺苷（cAMP），通过调节cAMP浓度，引发下游磷酸化级联反应，参与多种生理和病理过程。PDE4的作用机制是通过其催化活性中心，包含金属区和疏水区，特异性结合并水解cAMP，从而控制细胞内cAMP的水平。在生物学意义上，PDE4的活性调节与多种疾病密切相关，包括呼吸系统疾病、自身免疫性疾病、中枢神经系统疾病及皮肤病等，因此，PDE4抑制剂的开发为治疗这些疾病提供了新的策略。
