ADA,即腺苷脱氨酶,是一种参与嘌呤代谢途径的关键酶(EC3.5.4.4)。它在人体中的主要功能是免疫系统的发育和维持,尤其在淋巴细胞和巨噬细胞中发挥作用。ADA的作用机制包括不可逆地催化腺苷脱去氨基,转化为肌苷,然后肌苷可进一步被嘌呤核苷磷酸化酶(PNP)转化为次黄嘌呤。ADA活性位点含有锌离子,该离子是ADA活性所必需的唯一辅助因子,底物腺苷通过多个氢键与活性位点结合并被催化。ADA在信号传导中的作用涉及到调节细胞内腺苷水平,腺苷是一种强效的免疫调节分子,通过其受体影响细胞功能。ADA的活性还与多种生理和病理过程相关,包括上皮细胞分化、神经传递以及妊娠维持等。在生物学意义上,ADA不仅在嘌呤补救途径中发挥关键作用,而且其活性的变化与多种疾病相关,例如ADA缺乏可导致严重联合免疫缺陷(SCID),而ADA活性的增加与某些癌症、自身免疫疾病以及HIV/AIDS有关。因此,ADA是免疫调节和疾病诊断中的一个重要生物标志物。
