FSHR

卵泡刺激素受体(FSHR)属于G蛋白偶联受体超家族中的糖蛋白亚家族成员,由胞外区、胞内区及跨膜区3部分组成。FSHR通过与其配体——卵泡刺激素(FSH)结合参与信号转导。

FSH是垂体分泌的一种糖蛋白激素,主要作用是促进和维持正常的性腺发育和生殖功能。FSH可以通过与FSHR结合来刺激女性卵巢中卵泡的发育,促进男性睾丸的精子发生。FSH主要通过FSHR介导的腺苷酸环化酶-环磷酸腺苷(cAMP)途径发挥作用。已证实FSH通过特异性结合FSHR胞外区的富含亮氨酸重复序列来激活稳定态受体。活化的FSHR刺激位于细胞膜内膜的偶联鸟苷结合蛋白(Gs蛋白)传递信息。被活化的Gs蛋白激活腺苷酸化话梅促成胞质内第二信使cAMP生产,cAMP再激活蛋白激酶A,促进蛋白磷酸化,实现生理效应。

FSHR的表达被认为是性腺特异性的。长期以来,人们认为FSHR的表达仅限于卵巢的颗粒细胞和睾丸的支持细胞,但后来发现FSHR在胎盘、子宫、前列腺、骨组织和卵巢上皮细胞以及卵巢癌。此外,最近有研究发现FSHR在许多肿瘤血管表面选择性表达,并且发现FSHR的表达与原发性肾细胞癌的肿瘤转移密切相关。FSHR的表达水平在女性卵泡发育的不同阶段也不同。
