IL4R

IL-4R由802个氨基酸残基组成,分子量为140kDa,属于红细胞生成素受体超家族成员。其结构具有特定的特征,包括两个纤连蛋白结构域(fnIII),每个结构域由六个β片组成。在IL-4与IL-4Rα亚基结合时,这些结构域有助于形成异二聚体。IL-4R在多种细胞过程中扮演重要角色。它参与免疫细胞的存活、发育,调控B细胞的发育和成熟,以及Ⅱ型辅助性T细胞的增殖分化。此外,IL-4R的表达与多种肿瘤细胞的存活、增殖、迁移和侵袭密切相关。在肿瘤微环境中,IL-4R的表达上调,并通过多种机制促进肿瘤转移。 IL-4R的生物学功能主要通过非受体型酪氨酸激酶JAK激活。当IL-4与IL-4R结合时,它会激活JAK,进而触发一系列的信号级联反应。这些反应包括STAT6的磷酸化和核转位,以及其他下游信号分子的激活。这些信号通路最终影响细胞的增殖、分化、存活和迁移等过程。
