Ripk1

RIPK,即受体相互作用丝氨酸/苏氨酸蛋白激酶1,别名RIP1。RIPK1是一种胞质蛋白激酶,能够调节多种炎症和细胞死亡途径。它在肿瘤坏死因子受体1 (TNFR1) 信号通路中的作用已被广泛研究,其激酶活性对调控过程至关重要。RIPK1可以参与形成多种信号复合体,如复合体I(TNF-RSC),通过泛素化修饰促进NF-κB激活和抑制其激酶活性,从而抑制胚胎发生和炎症过程中RIPK1依赖的细胞死亡。RIPK1在生物学上的意义非常重大,它不仅参与细胞凋亡、坏死性凋亡(necroptosis)以及炎症信号通路的调控,还涉及胚胎发育、造血系统发育以及免疫稳态的维持等多种重要生物学过程。RIPK1的功能异常与多种疾病有关,包括自身免疫缺陷、自身炎症性疾病、渐冻症、风湿性关节炎以及结肠炎等。靶向RIPK1的小分子药物正在临床试验阶段,用于治疗这些疾病。此外,RIPK1的磷酸化、泛素化等翻译后修饰对其活性的调节在生理和病理条件下均发挥关键作用。

Immunofluorescence staining of HepG2 cells with CSB-PA618785LA01HU at 1:266, counter-stained with DAPI. The cells were fixed in 4% formaldehyde, permeabilized using 0.2% Triton X-100 and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4°C. The secondary antibody was Alexa Fluor 488-congugated AffiniPure Goat Anti-Rabbit IgG(H+L).
