SLC22A12

SLC22A12,即溶质载体家族22成员12,别名尿酸盐转运蛋白1(URAT1)、新型肾脏转运蛋白(NKT)。它是一种主要在肾脏近端小管上皮细胞的顶端刷缘膜上表达的跨膜蛋白,负责尿酸盐的重吸收过程。SLC22A12的作用机制是通过利用管腔两侧的浓度梯度和化学梯度差,将尿酸盐从近端肾小管腔内转运到肾小管上皮细胞,这是尿酸盐重吸收的第一步。SLC22A12在生物学上的意义主要体现在其对尿酸盐平衡的调控作用。它参与调节血尿酸水平,影响尿酸的排泄和重吸收。SLC22A12的功能缺失突变会导致尿酸盐重吸收功能障碍,可能引起1型肾低尿酸血症。此外,SLC22A12中的单核苷酸多态性(SNP)与代谢综合征的发病机制有关,包括肥胖、低高密度脂蛋白胆固醇、腰围增加等。因此,SLC22A12是研究和治疗与尿酸盐代谢相关的疾病,如痛风和高尿酸血症的潜在靶点。
