TRPV6

TRPV6,即瞬时受体电位通道家族,Vanilloid亚家族成员6”,别名ECACAC1。它是一种属于TRP阳离子通道家族的膜蛋白,主要在小肠、肾脏、胎盘、附睾和某些分泌腺等上皮组织中表达。TRPV6的作用机制涉及到对细胞内钙离子(Ca2+)的调节,通过其通道活性参与钙离子的选择性通透,对维持体内钙平衡和生理功能至关重要。TRPV6基因是维生素D活性形式的直接转录靶标,其表达受维生素D水平的调控,以适应机体对钙离子的需求。此外,TRPV6还可能通过与膜上的其他蛋白质如calmodulin相互作用来实现其功能。TRPV6在生物学上的意义主要体现在其对钙离子稳态的调控上。它在肠道和肾脏中的功能尤为重要,参与了钙离子的吸收和重吸收过程,从而帮助维持血钙水平。TRPV6的功能丧失突变与一些疾病相关,如高钙尿症、骨密度降低、体重增长不足、生育能力降低,甚至脱发。此外,TRPV6在一些恶性肿瘤中被发现过度表达,其上调与肿瘤的侵袭性、转移和不良生存率密切相关,表明TRPV6可能在肿瘤发展中起到促进作用。因此,TRPV6不仅在维持生理功能中发挥关键作用,也可能成为治疗某些疾病,尤其是癌症治疗的潜在靶点。

IHC image of CSB-PA861992LA01HU diluted at 1:300 and staining in paraffin-embedded human placenta tissue performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a biotinylated secondary antibody and visualized using an HRP conjugated SP system.
