脊髓灰质炎病毒受体4（Nectin-4）属于免疫球蛋白超家族（IgSF），是一类新发现的细胞黏附分子。NECTIN4作为一种I型膜蛋白，在正常的胚胎和胎儿组织中含量很高，成年后下降，在健康组织中的分布非常有限。然而，Nectin-4蛋白在尿路上皮癌、乳腺癌、膀胱癌、卵巢癌、胃癌、肝细胞癌和胰腺癌等组织中都具有特异性的高表达，其主要通过激活PI3K/Akt途径促进肿瘤细胞增殖、分化、迁移、侵袭等。目前，已上市的Nectin-4 ADC药物Padcev（Enfortumab vedotin）已展示出与免疫检查点抑制剂联用的优异临床潜力，用于局部晚期或转移性尿路上皮癌成人患者。未来，靶向Nectin-4的抗体-药物偶联物有望进一步将适应症拓宽至乳腺癌和胃癌等其它恶性肿瘤。
