Ddit3

DDIT3,即DNA损伤诱导转录本3,别名CHOP、GADD153,是一种转录因子,对细胞应激反应尤其是内质网应激具有关键调控作用。它通过与ATF4相互作用,促进自噬相关基因表达,并通过抑制mTOR信号通路激活细胞自噬。DDIT3还参与凋亡过程,通过调节Bcl-2家族和C/EBPδ诱导线粒体相关凋亡。DDIT3在肿瘤细胞自噬与凋亡调控中扮演重要角色,其异常表达可能增加肿瘤细胞对治疗的敏感性,是肿瘤治疗的潜在靶点。在谷氨酰胺饥饿条件下,DDIT3通过双重机制平衡糖酵解和线粒体氧化磷酸化,减少活性氧产生,帮助肿瘤细胞适应代谢压力,显示其在肿瘤细胞代谢调控中的生物学意义。

Then 10% normal goat serum to block non-specific protein-protein interactions followed by the antibody (1µg/1*106cells) for 45min at 4℃. The secondary antibody used was FITC-conjugated Goat Anti-rabbit IgG(H+L) at 1:200 dilution for 35min at 4℃.Control antibody (green line) was rabbit IgG (1µg/1*106cells) used under the same conditions. Acquisition of >10,000 events was performed.
