FAP

FAP,即成纤维细胞激活蛋白,别名Fibroblast Activation Protein-α、FAP-α。它是一种II型跨膜丝氨酸蛋白酶,通常在正常组织中表达量很低,但在纤维化、关节炎和癌症等病理性疾病中表达显著升高。FAP的作用机制包括促进肿瘤细胞的增殖、侵袭、血管生成、上皮向间质转化、干细胞促进、免疫抑制和耐药,其活性与肿瘤生长和转移密切相关。FAP通过与β-整合素结合,协同促进肿瘤细胞的增殖、迁移和侵袭,其促血管生成特性归因于它的二肽基肽酶活性,其作用底物神经肽Y在裂解后能促血管生成和促进内皮细胞迁移。FAP的下游信号通路主要包括PI3K/AKT、RAS/ERK、SHH/GLI、FAK等 。
