Ptk7

PTK7,即蛋白酪氨酸激酶7,别名CCK4(结肠癌激酶4)。它是一种跨膜受体蛋白,含有7个胞外Ig结构域、1个跨膜区、1个近膜区和1个催化惰性的细胞质酪氨酸激酶结构域。PTK7在脊椎动物的发育和胚胎生成中起着重要作用,特别是在平面细胞极化(PCP)和定向细胞运动的调节中。尽管PTK7缺乏酪氨酸激酶活性,但它能够与多种蛋白质相互作用,参与Wnt和非Wnt信号通路的调节,影响细胞的极性、运动、迁移和侵袭、组织再生和伤口愈合等过程。PTK7的作用机制涉及到其在Wnt/PCP信号通路和经典Wnt通路中的调节功能。在非典型Wnt通路中,PTK7能够与Frizzled受体和Dsh相互作用,参与信号的转导,激活GTP酶RhoA和Rac及其下游效应分子。在经典Wnt通路中,PTK7通过稳定细胞膜上的β-catenin发挥作用,促进LEF/TCF转录因子激活靶基因的表达。此外,PTK7还能够与VEGFR1相互作用,参与血管生成过程。PTK7的生物学意义主要体现在其对肿瘤发生、发展和转移的调控作用,其异常表达与多种肿瘤的发生和转移有关,尤其是在三阴性乳腺癌、非小细胞肺癌和卵巢癌中的表达量增加,与患者预后差有关。
