TNFRSF1B(TNF receptor superfamily member 1b)为TNF受体超家族成员,也称为TNFR2,主要表达在肿瘤微环境中的Treg细胞和多种肿瘤细胞中(如卵巢癌和肠癌)。TNFR2与受体TNF结合后,通过激活典型和非典型的NF-kB通路来控制细胞的生存和增殖。大量研究显示,TNFR2抑制剂能够特异性抑制在肿瘤微环境中高表达TNFR2的Tregs的功能,同时特异性杀伤肿瘤细胞,用于抗肿瘤治疗;而TNFR2激活剂则可以激活Tregs细胞,抑制效应性T细胞(Teff),进而下调免疫反应,可以用于自身免疫疾病治疗。随着对于新靶点的不断深入挖掘,TNFR2的重要性日渐受到关。TNFR2已经成为免疫逃逸、肿瘤生长以及检查点阻断抵抗性的潜在诱因。TNFR2抑制剂抗体则能够抑制肿瘤相关Tregs的增殖和功能。
