Erbb3

ERBB3(又称HER3,人表皮生长因子受体3)属于表皮生长因子受体家族(EGFR)。ERBB3在正常成人的胃肠道、生殖系统、皮肤、神经系统、泌尿道和内分泌系统中均有表达。事实上,ERBB3本身受到不同程度的调控,由2个泛素连接酶NEDD4和NRDP1调控其泛素化和蛋白体降解。ERBB3胞内区无内在的酪氨酸激酶活性不能自身磷酸化,但ERBB3能够与表皮生长因子受体家族其它成员,ERBB1(又称EGFR,HER1)、ERBB2/HER2和ERBB4/HER4形成异源性二聚体,激活下游通路,影响细胞的增殖分化。其中ERBB3/ERBB2复合物能够激活MAPK/ERK和PI3K/AKT在内的多种通路。大量研究表明, ERBB3过表达可见于乳腺癌、肺癌、结直肠癌、胃癌、头颈鳞癌、卵巢癌、黑色素瘤、前列腺癌和膀胱癌等。因此,通过靶向ERBB3,阻断ERBB3/ERBB2通路,正在为癌症治疗提供新途径。
